Jing Zhang, Xiu Han, Qun Lu, Yunfei Feng, Aiqun Ma & Tingzhong Wang
Abstract Left ventricular non-compaction cardiomyopathy (LVNC) is one of the most common inherited cardiovascular diseases. The genetic backgrounds of most LVNC patients are not fully understood. We collected clinical data, family histories, and blood samples and performed genetic analysis using next-generation sequencing (NGS) from a Chinese family of 15 subjects. Clinically LVNC affected subjects showed marked cardiac phenotype heterogeneity. We found that these subjects with LVNC carried a missense heterozygous genetic mutation c.905G>A (p.R302Q) in...

Screening of differentially expressed microRNAs and target genes in two potato varieties under nitrogen stress

Yue Lu, Jingying Zhang, Zhijun Han, Zhongcai Han, Shuang Li, Jiayue Zhang, Haoran Ma & Yuzhu Han
Abstract Background A reasonable supply of nitrogen (N) fertilizer is essential for obtaining high-quality, high-level, and stable potato yields, and an improvement in the N utilization efficiency can effectively reduce N fertilizer use. It is important to use accurate, straightforward, and efficient transgenic breeding techniques for the identification of genes that can improve nitrogen use efficiency, thus enabling us to achieve the ultimate goal of breeding N-efficient potato varieties. In recent years, some of the...

Construction of a Carcinoembryonic Antigen Surface-Enhanced Raman Spectroscopy (SERS) Aptamer Sensor Based on the Silver Nanorod Array Chip

Rui Li, Lijun Li, Yan Zhang, Xin Lin, Heyuanxi Guo, Chubing Lin & Jun Feng
Carcinoembryonic antigen (CEA) is a cancer-related tumor marker, which is commonly used for preventive screening, auxiliary diagnosis, and recurrence monitoring. Therefore, it is of great significance to develop a new CEA detection method. In this paper, we developed an SERS aptasensor for CEA based on silver nanorod array chip, thiol aptamer, and 4-mercaptophenylboronic acid (4-MPBA). The silver nanorod array chip modified by CEA thiol aptamer (aptamer-SH) was used as SERS capture substrates. Ag@4-MPBA was used...

USP38 inhibits colorectal cancer cell proliferation and migration via downregulating HMX3 ubiquitylation

Jun Wang, Yongxing Gu, Xueqin Yan, Jie Zhang & Yong Ding
Accumulating evidence has shown that H6 Family Homeobox 3 (HMX3) plays a crucial role in nervous system regulation. However, the regulatory mechanism of HMX3 in colorectal cancer (CRC) has seldom been studied. Herein, HMX3 was significantly downregulated in CRC, as demonstrated by qRT-PCR and WB analysis on clinical samples and a panel of cell lines. Besides, it was found that the expression of HMX3 was negatively correlated with survival of CRC patients. The functional analyses...

Overcoming high level adenosine-mediated immunosuppression by DZD2269, a potent and selective A2aR antagonist

Yu Bai, Xin Zhang, Jie Zheng, Ziyi Liu, Zhenfan Yang & Xiaolin Zhang
Abstract Background Adenosine is a potent immunosuppressant whose levels in the tumor microenvironment (TME) are often much higher than those in normal tissues. Binding of adenosine to its receptor A2aR activates a cascade of genes and leads to immunosuppression. In addition, immune checkpoint blockage markedly increases A2aR expression in T cells, which could dampen their anti-tumor response. Several A2aR antagonists are under clinical development, but with limited clinical benefit reported so far. These A2aR antagonists...

Activation function 1 of progesterone receptor is required for progesterone antagonism of oestrogen action in the uterus

Shi Hao Lee, Chew Leng Lim, Wei Shen, Samuel Ming Xuan Tan, Amanda Rui En Woo, Yeannie H. Y. Yap, Caitlyn Ang Su Sian, Wilson Wen Bin Goh, Wei-Ping Yu, Li Li & Valerie C. L. Lin
Abstract Background Progesterone receptor (PGR) is a master regulator of uterine function through antagonistic and synergistic interplays with oestrogen receptors. PGR action is primarily mediated by activation functions AF1 and AF2, but their physiological significance is unknown. Results We report the first study of AF1 function in mice. The AF1 mutant mice are infertile with impaired implantation and decidualization. This is associated with a delay in the cessation of epithelial proliferation and in the initiation...

Hepatitis B virus promotes hepatocellular carcinoma development by activating GP73 to repress the innate immune response

Long Liu, Yanping Huang, Yanan Fu, Jingjing Rao, Feng Zeng, Manshan Ji, Xiang Xu, Jianyong Zhu, Weixing Du & Zhixin Liu
Abstract Background Hepatitis B virus (HBV) causes acute and chronic infection in the clinic. Hepatocellular carcinoma (HCC) is closely linked to HBV infection. Serum Golgi protein 73 (GP73) increases during HBV infection. However, the role of GP73 during HBV infection and the occurrence of HBV-related HCC is still poorly understood. Methods The underlying role of HBV-induced GP73 in regulating HCC development was investigated in this study. GP73 expression in HBV-related clinical HCC tissues and in...

Host plants affect the susceptibility of Spodoptera exigua to its homologous nucleopolyhedrovirus: role of chitin synthase and intestinal mucin in the peritrophic matrix

Host plants have an effect on the susceptibility of lepidopteran insects to nucleopolyhedrovirus (NPV), but the role of the protein in peritrophic membrane (PM) of larvae has not been investigated. We determined a key PM gene (SeIIM8 gene) in Spodoptera exigua (Hübner), which plays a role in protecting against NPV infection. Then we quantified the activity of chitin synthase (SeCHS), the content of insect intestinal mucin (SeIIM) and the expression of their genes in S....

Preparation and application of carboxyalkyl-modified organosilicon compounds

Yangchao Cheng, Yuxia Zhang, Xue Hu, Hong Dong, Zhirong Qu, Xuyang Cheng, Tao Zhang, Wei Chen, Pengchun Shi & Chuan Wu
Hydroxyalkyl-functionalized organosilicon compounds have been prepared from hydrogen-containing organosilicon compounds by hydrosilylation. Various carboxyalkyl-modified organosilicon compounds were then prepared by reacting the hydroxyalkyl-functionalized organosilicon compounds with various cyclic acid anhydrides under solvent and catalyst free. The effects of the nature of the hydroxyalkyl functional groups and the structure of the organosilicon compounds on the ring-opening reaction have been investigated. The synthesized carboxyalkyl-modified organosilicon compounds have been used to treat cotton fabric samples, and the effects...
