Characterization of key aroma compounds and regulation mechanism of aroma formation in local Binzi (Malus pumila × Malus asiatica) fruit

Qinghua Wang, Fan Gao, Xuexue Chen, Wenjiang Wu, Lei Wang, Jiangli Shi, Yun Huang, Yuanyue Shen, Guoliang Wu & Jiaxuan Guo
Abstract Background Volatile components are important secondary metabolites essential to fruit aroma quality, thus, in the past decades many studies have been extensively performed in clarifying fruit aroma formation. However, aroma components and biosynthesis in the fruit of Binzi (Malus pumila × Malus asiatica), an old local species with attractive aroma remain unknown. Results We investigated two Binzi cultivars, ‘Xiangbinzi’ (here named high-fragrant Binzi, ‘HFBZ’) and ‘Hulabin’ (here named low-fragrant Binzi, ‘LFBZ’) by monitoring the...

TET2 regulates osteoclastogenesis by modulating autophagy in OVX-induced bone loss

Chen Yang, Huaqiang Tao, Haifeng Zhang, Yu Xia, Jiaxiang Bai, Gaoran Ge, Wenming Li, Wei Zhang, Long Xiao, Yaozeng Xu, Zhirong Wang, Ye Gu, Huilin Yang, Yu Liu & Dechun Geng
Increased bone resorption by osteoclasts after estrogen deficiency is the main cause of postmenopausal osteoporosis. TET2 (tet methylcytosine dioxygenase 2) is a DNA demethylase that regulates cellular function and differentiation potential. Macroautophagy/autophagy maintains cellular homeostasis by recycling unnecessary and damaged organelles. This study revealed that TET2 promoted bone loss in oophorectomized (OVX) mice and that TET2 promoted osteoclast differentiation by regulating autophagy. Tet2 knockdown inhibited autophagy and osteoclast differentiation in vitro. Mechanistically, Tet2 knockdown increased...

Nanoparticle-mediated delivery of Tanshinone IIA reduces adverse cardiac remodeling following myocardial infarctions in a mice model: role of NF-κB pathway

Shuai Mao, Lei Wang, Peipei Chen, Yong Lan, Rui Guo & Minzhou Zhang
Our previous works have shown that tanshinone IIA inhibited maladaptive extracellular matrix remodeling in cardiac fibroblasts implicating its potential role in treating of pathologic cardiac remodeling. However, the intrinsically poor solubility and bioavailability of tanshinone IIA hindered its clinical application. Here we develop monomethoxy-poly (ethylene glycol)-poly (lactic acid)-D-α-Tocopheryl polyethylene glycol 1000 succinate (mPEG-PLA-TPGS) nanoparticle incorporating tanshinone IIA (tanshinone IIA-NPs) and study its efficacy in post-infarction left ventricular (LV) remodeling. Male C57BL/6 mice underwent left coronary...

Morphometric and Hemodynamic Analysis of the Compressed Iliac Vein

Hongyu Wang, Wei Jia, Yifeng Xi, Yuan Li, Yubo Fan, Xiaoyan Deng & Zengsheng Chen
Purpose:To investigate the relationship between the morphological structure and hemodynamic properties of the compressed iliac vein and explore the reason for the formation of thrombosis in the compressed iliac vein.Materials and Methods:A total of 11 patients with iliac vein compression syndrome (IVCS) were included in this study, and their iliac veins were reconstructed in 3 dimensions (3D). The morphological structures of the iliac veins (confluence angle, degree of stenosis) were analyzed based on the 3D...

LINC00305 represses miR-124 expression to trigger inflammatory insults in the presence of lipopolysaccharide

Xiaoyan Li, Manqiu Yu, Liang Han, Lei Chen, Daxue Zhang, Guannan Zhou, Qiang Zhao & Taitao Sun
The anti-inflammatory function of microRNA-124 (miR-124) has been a matter of extensive studies in the last few years. Although LINC00305 regulates biological activities by acting as a miR sponge, it is still unexplored whether LINC00305 is involved in inflammation by regulating miR-124. Cell viability and apoptosis were evaluated with commercial kits, cell counting kit-8 (CCK-8) and Annexin V-fluorescein isothiocyanate (FITC) kit, respectively. LINC00305, miR-124 and mRNA levels were quantified by quantitative reverse transcription PCR (qRT-PCR)....

MiR-30a-5p frequently downregulated in prostate cancer inhibits cell proliferation via targeting PCLAF

Hu Zhao, Wei Zhang, Xiaofeng Lai, Hehuan Zhu, Shenhang Zhang, Weizhen Wu, Shuiliang Wang, Minying Tang, Zhen Deng & Jianming Tan
MicroRNAs (miRNAs) have vastly expanded our view of RNA world in intracellular signal regulating networks. Here, we functionally characterized a normally highly expressed miRNA, miR-30a-5p (MIMAT0000087), which exhibits downregulated expression profiles in prostate cancer samples. MiR-30a-5p knockdown and overexpression in PC-3 cell line alters cell proliferation supporting a tumour suppressor role. We also discovered that PCLAF is the direct target of miR-30a-5p. Notably, PC-3 cell proliferation is inhibited by miR-30a-5p/PCLAF axis. miR-30a-5p represents a novel...

β-galactosidase GALA from Bacillus circulans with high transgalactosylation activity

Yaru Yan, Weishi Guan, Xiaoyi Li, Kaier Gao, Xinxin Xu, Bo Liu, Wei Zhang & Yuhong Zhang
β-galactosidase catalyzes lactose hydrolysis and transfers reactions to produce prebiotics such as galacto-oligosaccharides (GOS) with potential applications in the food industry and pharmaceuticals. However, there is still a need for improved transgalactosylation activity of β-galactosidases and reaction conditions of GOS production in order to maximize GOS output and reduce production costs. In this study, a β-galactosidase gene, galA, from Bacillus circulans was expressed in Pichia pastoris, which not only hydrolyzed lactose but also had strong...

POU class 2 homeobox associating factor 1 (POU2AF1) participates in abdominal aortic aneurysm enlargement based on integrated bioinformatics analysis

Jinze Meng, Hao Wen, Xintong Li, Boyang Luan, Shiqiang Gong, Jie Wen, Yifei Wang & Lei Wang
Abdominal aortic aneurysm (AAA) is life-threatening, its natural course is progressively sac expansion and rupture. Elegant studies have been conducted to investigate the molecular markers associated with AAA growth and expansion, this topic however, still needs to be further elucidated. This study aimed to identify potential genes for AAA growth and expansion based on comprehensive bioinformatics approaches. Firstly, 29 up-regulated genes were identified through DEGs analysis between large AAA and small AAA in GSE57691. Secondly,...
