Autophagy and its mediated mitochondrial quality control maintain pollen tube growth and male fertility in Arabidopsis
He Yan, Menglong Zhuang, Xiaoyu Xu, Shanshan Li, Mingkang Yang, Nianle Li, Xiaojuan Du, Kangwei Hu, Xiaomin Peng, Wei Huang, Hong Wu, Yu Chung Tse, Lifeng Zhao & Hao Wang
Macroautophagy/autophagy, a major catabolic pathway in eukaryotes, participates in plant sexual reproduction including the processes of male gametogenesis and the self-incompatibility response. Rapid pollen tube growth is another essential reproductive process that is metabolically highly demanding to drive the vigorous cell growth for delivery of male gametes for fertilization in angiosperms. Whether and how autophagy operates to maintain the homeostasis of pollen tubes remains unknown. Here, we provide evidence that autophagy is elevated in growing...
SDC1-dependent TGM2 determines radiosensitivity in glioblastoma by coordinating EPG5-mediated fusion of autophagosomes with lysosomes
Wang Zheng, Qianping Chen, Hongxia Liu, Liang Zeng, Yuchuan Zhou, Xinglong Liu, Yang Bai, Jianghong Zhang, Yan Pan & Chunlin Shao
Glioblastoma multiforme (GBM) is the most common brain malignancy insensitive to radiotherapy (RT). Although macroautophagy/autophagy was reported to be a fundamental factor prolonging the survival of tumors under radiotherapeutic stress, the autophagic biomarkers coordinated to radioresistance of GBM are still lacking in clinical practice. Here we established radioresistant GBM cells and identified their protein profiles using tandem mass tag (TMT) quantitative proteomic analysis. It was found that SDC1 and TGM2 proteins were overexpressed in radioresistant...
Transcriptome analysis of adipocytokines and their-related LncRNAs in lung adenocarcinoma revealing the association with prognosis, immune infiltration, and metabolic characteristics
Jie Ren, Hui Zhang, Jinna Wang, Yingsong Xu, Lei Zhao & Qihang Yuan
Lung adenocarcinoma (LUAD) is amongst the major contributors to cancer-related deaths on a global scale. Adipocytokines and long non-coding RNAs (lncRNAs) are indispensable participants in cancer. We performed a pan-cancer analysis of the mRNA expression, single nucleotide variation, copy number variation, and prognostic value of adipocytokines. LUAD samples were obtained from the Gene Expression Omnibus (GEO) and The Cancer Genome Atlas (TCGA) databases. Simultaneously, train, internal and external cohorts were grouped. After a stepwise screening...
Prevalence of major morbidities and outcome of all hospitalized neonates. A retrospective cohort study of Huai’an neonatal survivals
Yaling Xu, Xiaoqin Zhu, Hui Wang, Zhaojun Pan, Xiaoqiong Li, Xiaojing Guo, Hongni Yue & Bo Sun
Reliable data for causal implication of neonatal survival in China are lacking. We assumed that by analyzing surviving data of in-hospital neonatal care based on all livebirths in Huai’an, prevalence of neonatal morbidity, mortality and causal relations may be derived comprehensively. Data of all regionally hospitalized neonates were retrospectively linked with corresponding whole livebirths (59,056) in 2015 as a cohort. Diagnoses of diseases and causes of deaths were redefined and categorized. Disease patterns, prevalence of...

Interleukin-17A attenuates photoreceptor cell apoptosis in streptozotocin-induced diabetic mouse model
Minqi Zhu, Shuang Gao, Sha Gao, Yanuo Wang, Na Li & Xi Shen
Diabetic retinopathy (DR) represents an important microvascular complication of diabetes, which is the top etiology of vision impairment worldwide. Although interleukin (IL)-17A is increasingly implicated in DR development, the underlying cellular mechanisms remain poorly defined. This work aims to evaluate IL-17A levels in the retina of streptozotocin (STZ)-induced diabetic mice and elucidate their potential roles. We found IL-17A was upregulated in diabetic retina after intraperitoneal injection of STZ and high-glucose (HG)-cultured primary Müller cells. IL-17A...

Bayesian Optimal Phase II Design for Randomized Clinical Trials
Yujie Zhao, Bo Yang, J. Jack Lee, Li Wang & Ying Yuan
Randomized clinical trials are the gold standard to evaluate the efficacy of an experimental treatment. We propose a flexible Bayesian optimal phase II (BOP2) design for two-arm randomized trials. The proposed two-arm BOP2 design is flexible and can handle single, multiple primary and coprimary endpoints for superiority and noninferiority trials under a unified framework. It also allows users to specify the number and timing of interim analyses to meet clinical needs. While enjoying the flexibility...
