Phenylboronic ester-modified polymeric nanoparticles for promoting TRP2 peptide antigen delivery in cancer immunotherapy
Qiyan Wang, Zhipeng Dong, Fangning Lou, Yunxue Yin, Jiahao Zhang, Hanning Wen, Tao Lu & Yue Wang
The tremendous development of peptide-based cancer vaccine has attracted incremental interest as a powerful approach in cancer management, prevention and treatment. As successful as tumor vaccine has been, major challenges associated with achieving efficient immune response against cancer are (1) drainage to and retention in lymph nodes; (2) uptake by dendritic cells (DCs); (3) activation of DCs. In order to overcome these barriers, here we construct PBE-modified TRP2 nanovaccine, which comprises TRP2 peptide tumor antigen...
Regulation of TFEB nuclear localization by HSP90AA1 promotes autophagy and longevity

TFEB (transcription factor EB) regulates multiple genes involved in the process of macroautophagy/autophagy and plays a critical role in lifespan determination. However, the detailed mechanisms that regulate TFEB activity are not fully clear. In this study, we identified a role for HSP90AA1 in modulating TFEB. HSP90AA1 was phosphorylated by CDK5 at Ser 595 under basal condition. This phosphorylation inhibited HSP90AA1, disrupted its binding to TFEB, and impeded TFEB’s nuclear localization and subsequent autophagy induction. Pro-autophagy...

A population pharmacokinetic-pharmacodynamic model of navtemadlin, its glucuronide metabolite (M1) and serum macrophage inhibitory cykokine-1 (MIC-1)
Lu Zhang, Bill Poland, Michelle Green, Shekman Wong & J. Greg Slatter
Navtemadlin is a potent, selective, orally available inhibitor of murine double minute 2 that restores p53 activity to induce apoptosis in TP53 wild-type malignancies. Using richly sampled pharmacokinetic (PK) and pharmacodynamic (PD) data from healthy volunteers, a population PK/PD model was developed. A population PK (PPK) model described the PK characteristics of navtemadlin and its major metabolite acyl glucuronide (M1) and quantified enterohepatic recirculation (EHR). Post hoc individual PK parameters from this model were coupled...

Pediatric and adult asthma clinical phenotypes: a real world, big data study based on acute exacerbations
Jie Xu, Jiang Bian & Jennifer N Fishe
Asthma is a heterogeneous disease with a range of observable phenotypes. To date, the characterization of asthma phenotypes is mostly limited to allergic versus non-allergic disease. Therefore, the aim of this big data study was to computationally derive asthma subtypes from the OneFlorida Clinical Research Consortium We obtained data from 2012-2020 from the OneFlorida Clinical Research Consortium. Longitudinal data for patients greater than two years of age who met inclusion criteria for an asthma exacerbation...

For each pairwise comparison, the functional enrichments in gene ontologies of the biological processes (A), cellular components (B), and molecular functions (C) categories were tested for the differentially expressed genes up- or down-regulated (UP, and DOWN, respectively). The labels of pairwise comparisons were coded in two letters: the first one represents the phenotype (M: mothers, F: fathers, P: paternal males,...
