Inhibition of CDK9 activity compromises global splicing in prostate cancer cells
Qiang Hu, Ninu Poulose, Samuel Girmay, Alma Helevä, Dimitrios Doultsinos, Aishwarya Gondane, Rebecca E Steele, Xiaozhuo Liu, Massimo Loda, Song Liu, Dean G. Tang, Ian G Mills & Harri M Itkonen
Cyclin-dependent kinase 9 (CDK9) phosphorylates RNA polymerase II to promote productive transcription elongation. Here we show that short-term CDK9 inhibition affects the splicing of thousands of mRNAs. CDK9 inhibition impairs global splicing and there is no evidence for a coordinated response between the alternative splicing and the overall transcriptome. Alternative splicing is a feature of aggressive prostate cancer (CRPC) and enables the generation of the anti-androgen resistant version of the ligand-independent androgen receptor, AR-v7. We...

Foeniculins A-C (1–3) together with a pair of enantiomers (±)-foeniculin D (4) were isolated from endophytic fungus Diaporthe foeniculina BZM-15. Their structures including absolute configurations were unambiguously established by extensive interpretation of the NMR and HR-ESI-MS data, ECD measurements powered by molecular calculations, as well as Mo2(OAc)4 mediated CD methodology. The cytotoxic activity assay disclosed that these compounds didn’t show any noticeable cytotoxic activity.

Hang Huang, Haokai Xu, Ping Li, Xueting Ye, Wei Chen & Xixi Huang
The present study was aimed to evaluate the expression profile of Zinc finger C3H1 domain-containing protein (ZFC3H1) using bioinformatic analysis of public datasets from The Cancer Genome Atlas database (TCGA). The results showed that the expression levels of ZFC3H1 were notably lower than the corresponding non-cancerous tissues in prostate adenocarcinoma (PRAD), and patients in the high ZFC3H1-expression group showed poor survival. We hypothesized that the low expression of ZFC3H1 in tumor tissue might have be...

Patients with depression have a high prevalence of developing dyslipidemia. In this study, we aim to investigate the difference of serum lipids, including total cholesterol (TCH), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), and triglycerides (TG), between the depressed patients and healthy controls. Sex differences in lipids and their psychological correlations were also included. The study included 56 healthy controls (males/females =26/30) and 110 first-diagnosed drug-naïve outpatients (males/females =35/75). A total of 42 patients...

A growing body of evidence suggests the pivotal role of long non-coding RNA (lncRNA) in influenza virus infection. Based on next-generation sequencing, we previously demonstrated that Lnc45 was distinctively stimulated by H5N1 influenza virus in mice. In this study, we systematically investigated the specific role of Lnc45 during influenza A virus (IAV) infection. Through qRT-PCR, we first demonstrated that Lnc45 is highly up-regulated by different subtypes of IAV strains, including H5N1, H7N9, and H1N1 viruses....

Jing Chen, Li-Na Huo, Yuan Gao, Yun-Li Zhang & Ye Chen
Two new N-acetyl-ᴅ-glucosamine derivatives, penichryfurans A (1) and B (2), were obtained from the fermentation of an endophytic fungus Penicillium chrysogenum which inhabited the marine medical red alga Grateloupia turuturu. Their structures were established by detailed spectroscopic analysis of 1D and 2D NMR in combination with HRESIMS data. The absolute configurations were determined by the (Mo2(OAc)4)-induced CD and comparison of the calculated and experimental electronic curcular dichroism (ECD) spectra. Both compounds were evaluated for their...
Ferritinophagy is involved in the zinc oxide nanoparticles-induced ferroptosis of vascular endothelial cells
Xia Qin, Jun Zhang, Bin Wang, Ge Xu, Xi Yang, Zhen Zou & Chao Yu
Zinc oxide nanoparticles (ZnONPs) hold great promise for biomedical applications. Previous studies have revealed that ZnONPs exposure can induce toxicity in endothelial cells, but the underlying mechanisms have not been fully elucidated. In this study, we report that ZnONPs can induce ferroptosis of both HUVECs and EA.hy926 cells, as evidenced by the elevation of intracellular iron levels, lipid peroxidation and cell death in a dose- and time-dependent manner. In addition, both the lipid reactive oxygen...

Wenwen Zhu, Jia Wang, Na Zhao, Rui Zheng, Dalu Wang, Weiwei Liu & Beixing Liu
Changes in the intestinal microbiota indirectly impact the health of mucosa distal to the intestine, particularly the respiratory tract. However, the effects of intestinal microbiota dysbiosis on the regulation of respiratory syncytial virus (RSV) infection are not clear. In this study, we examined the effects of altering the intestinal microbiota on the pulmonary immune response against RSV infection. BALB/c mice were treated with streptomycin before infection with RSV to study the altered immune response. The...
