Synthesis, crystal structure and coordination behaviors of two unexpected tetranuclear Zn(II) and Co(II) supramolecular boxes derived from structural variation of coumarin schiff base ligand
Yin-Xia Sun, Juan Li, Jin-Guo Li, Zhe-Peng Deng, Yu Sun & Li Xu
Two unexpected tetranuclear supramolecular boxes, [Zn4(L')4(CH3OH)2]·2CH3OH (1) and [Co4(L)2(L')2(CH3OH)2]·acetone (2), with M4(μ3-O)2(μ2-O)4 core were obtained through reacting coumarin Schiff base H2L (H2L = 4-hydroxy-3-((2-hydroxyphenyl-imino)-methyl)-2hchrome-2-one) with Zn(OAc)2·4H2O and Co(OAc)2·4H2O, respectively. Interestingly, the original ligand (H2L) has changed into a new structure (H2L') in the process of forming single crystals. The structures of 1 and 2 were characterized by X-ray single crystal diffraction. 1 and 2 are tetranuclear structures including four metal centers (Zn(II) or Co(II)), four...

The non-receptor protein tyrosine phosphatase (PTP) SHP2 encoded by the PTPN11 gene is a critical regulator in a number of cellular signalling processes and pathways, including the MAPK and the immune-inhibitory programmed cell death PD-L1/PD-1 pathway. Hyperactivation and inactivation of SHP2 is of great therapeutic interest for its association with multiple developmental disorders and cancer-related diseases. In this work, we characterised a potent SHP2 allosteric inhibitor 2-((3 R,4R)-4-amino-3-methyl-2-oxa-8-azaspiro[4.5]decan-8-yl)-5-(2,3-dichlorophenyl)-3-methylpyrrolo[2,1-f][1,2,4]triazin-4(3H)-one (PB17-026-01) by using structure-based design. FOXP3+ macrophage represses acute ischemic stroke-induced neural inflammation
Wei Cai, Mengyan Hu, Chunyi Li, Ruizhen Wu, Danli Lu, Chichu Xie, Wei Zhang, Tiemei Li, Shishi Shen, Huipeng Huang, Wei Qiu, Quentin Liu, Yan Lu & Zhengqi Lu
Proper termination of cell-death-induced neural inflammation is the premise of tissue repair in acute ischemic stroke (AIS). Macrophages scavenge cell corpses/debris and produce inflammatory mediators that orchestrate immune responses. Here, we report that FOXP3, the key immune-repressive transcription factor of Tregs, is conditionally expressed in macrophages in stroke lesion. FOXP3 ablation in macrophages results in detrimental stroke outcomes, emphasizing the beneficial role of FOXP3+ macrophages. FOXP3+ macrophages are distinct from the M1 or M2 subsets...

Negative Regulation of RIG-I by Tim-3 Promotes H1N1 Infection
Qingzhu Shi, Ge Li, Shuaijie Dou, Lili Tang, Chunmei Hou, Zhiding Wang, Yang Gao, Zhenfang Gao, Ying Hao, Rongliang Mo, Beifen Shen, Renxi Wang, Yuxiang Li & Gencheng Han
The mechanisms by which retinoic acid-inducible gene I (RIG-I), a critical RNA virus sensor, is regulated in many biological and pathological processes remain to be determined. Here, we demonstrate that T cell immunoglobulin and mucin protein-3 (Tim-3), an immune checkpoint inhibitor, mediates infection tolerance by suppressing RIG-I-type I interferon pathway. Overexpression or blockade of Tim-3 affects type I interferon expression, virus replication, and tissue damage in mice following H1N1 infection. Tim-3 signaling decreases RIG-I transcription...
Sclerostin is involved in osteogenic transdifferentiation of vascular smooth muscle cells in chronic kidney disease-associated vascular calcification with non-canonical Wnt signaling
Qiong Xiao, Yun Tang, Haojun Luo, Sipei Chen, Rong Chen, Zhe Yan, Lei Pu, Li Wang, Guisen Li & Yi Li
Vascular calcification is prominent in patients with chronic kidney disease (CKD) and is a strong predictor of cardiovascular mortality in the CKD population. However, the mechanism underlying CKD-associated vascular calcification remains unclear. To identify potential therapeutic targets, a 5/6 nephrectomy rat model was established by feeding of a high-phosphorous diet as the CKD group and compared with sham group rats at 4 and 16 weeks. Sequencing analyses of the rat aorta revealed 643 upregulated and...
